    HSF Family Introduction

    During the initial stages of heat stress, the heat shock transcription factor (HSF) is the primary molecule responsible for relaying signals of cellular stress to the transcriptional apparatus. Metazoan HSFs comprise a rather small gene family consisting of four distinct types. In vertebrates, HSF1 is considered to be the predominant heat-inducible HSF, while HSF2 responds to the developmental signals and is strongly activated in the erythroleukemia cells by hemin treatment (Sistonen et al., 1992, 1994; Baler et al., 1993; Sarge et al., 1993). Avian species have an additional factor, HSF3, that functions during heat stress and is cell type-specific (Nakai and Morimoto, 1993; Nakai, et al., 1995). In addition to HSFs that activate transcription, HSF4 in man has no capacity to support activated transcription and has been shown to suppress expression of heat shock genes under control conditions (Nakai et al., 1997).

    Based on parsimony analysis of their DNA-binding domains (DBDs) and a comparison of oligomerization domains (ODs), they have been assigned to major classes A and B (Czarnecka-Verner et al., 1995, 1997, ; Nover et al., 1996; Nover and Scharf, 1997; Czarnecka-Verner and Gurley, 1999).These two groups of HSFs appear to be unique to plants with no clear relationships to HSF types in other organisms. Other differences between class A and B HSFs include the relative position of the nuclear localization sequence (NLS), and the presence or absence of a C-terminal hydrophobic region (HR-C), which in vertebrates has been shown to be involved in regulation of the trimerization state of HSF. In addition, the class B HSFs have much shortened C-terminal domains and, with the exception of tomato LpHSFB1 (LpHSF24), seem to lack functional transcriptional activation motifs (AHA or TRP repeats) which are present in class A HSFs.
